What is MK-677?
Unlike injectable GHRPs, MK-677 is orally bioavailable with a once-daily profile, elevating GH pulsatility and IGF-1 for 24 hours. Human studies have examined muscle wasting, bone density, sleep architecture (it increases REM and deep sleep duration) and growth hormone deficiency. Increased appetite - the ghrelin signature - is its most consistent effect. It is the reference oral secretagogue against which the injectable peptide class is compared.
Mechanism of Action (Research Context)
Agonist at the ghrelin/growth hormone secretagogue receptor (GHS-R1a) in pituitary and hypothalamus, amplifying pulsatile GH release and downstream hepatic IGF-1 production. Oral bioavailability and long functional duration distinguish it from peptide GHRPs.
