ACE-031 is a growth factor research compound with a molecular weight of ~90-100 kDa (dimer) and a research half-life of ~10-14 days (fc-fusion). ACE-031 is a recombinant fusion protein combining the extracellular domain of the activin receptor type IIB (ActRIIB) with the Fc region of human IgG1. It acts as a soluble decoy receptor, capturing myostatin, activins and other TGF-beta ligands before they can signal through cell-surface receptors. It was developed by Acceleron Pharma and reached Phase 2 trials in Duchenne muscular dystrophy before development was halted.

What is ACE-031?

In clinical research ACE-031 produced clear increases in lean body mass and thigh muscle volume after a single dose, validating the ligand-trap approach to myostatin pathway inhibition. The Fc fusion gives it an antibody-like half-life of around two weeks - completely unlike short-lived peptides. It remains an important reference compound in muscle-wasting research, and its trial data is frequently cited when evaluating newer ActRIIB pathway agents such as bimagrumab.

Mechanism of Action (Research Context)

Soluble ligand trap: the ActRIIB extracellular domain binds myostatin (GDF-8), activins A/B and GDF-11 with high affinity, preventing ActRIIB/ALK4-5 receptor activation and SMAD2/3 signaling. The IgG1 Fc domain confers extended circulation via FcRn recycling.
